Masuku was Johannesburg's Member of the Mayoral Committee for Finance and deputy ANC regional secretary before winning the ANC's 16th Johannesburg Regional Elective Conference in December 2025, beating Dada Morero by 184 votes to 149 to become regional chairperson, the first woman in that post according to Sunday World.
